He and his wife Mary lived in New London, New London Co., Connecticut.3 On 1 September 1722 his children relinquished their rights to his estate to their uncle Daniel Comstock.3 However, seemingly contradicting this, on 3 March 1739/40 his heirs agreed to the distribution of his estate, with 1/3 going to his widow Mary. Of the remainder, 2/7 went to the children of his son Kingsland, and 1/7 each to his children Daniel, Benjamin, Mary, and Ann, and 1/7 to his deceased son Joseph's daughter Sarah.3

He and his wife Elizabeth lived in Norwalk, Fairfield Co., Connecticut.2 On 6 November 1694 the court approved the inventory of his estate, which was valued at £353 16s.2 He and Elizabeth had just one child, Daniel (b 1693, d 5 March 1782).4

He served as deputy to the General Assembly in 1711 and 1714, and from 1726 to 1730.2 He also served in the South Company of militia in Norwalk, holding ranks of Ensign, Lieutenant, and Captain.2

He lived in Norwalk, Fairfield Co., Connecticut.2 On 14 March 1705/6 he received a grant of 1.5 acres on Long Island (in the Housatonic River) in Fairfield Co., Connecticut.2 On 22 May 1707 he and his mother Hannah deeded to his brother Samuel part of his father Christopher's home lot.2 On 11 December 1749 he deeded land at Silver Mine Hill to his son Moses.2
On 2 February 1725/26 he was appointed guardian of his nephews Samuel and James Olmstead, presumably sons of his sister Mercy.2
In his book Letters of a Traveller, published in 1850, William Cullen Bryant describes a conversation he had with a "robust old gentleman," who was a relative and friend of Moses Comstock's spinster daughters Sarah and Phebe. In describing their father Moses, Bryant wrote:
He prided himself on the strict and literal fulfillment of his word. On one occasion he had a law-suit with one of his neighbors, before a justice of the peace, in which he was cast and ordered to pay ten shillings damages, and a shilling as the fees of the court. He paid the ten shillings, and asked the justice whether he would allow him to pay the remaining shilling when he next passed his door. The magistrate readily consented, but from that time old Comstock never went by his house. Whenever he had occasion to go to church, or any other place, the direct road to which led by the justice's door, he was careful to take a lane which passed behind the dwelling, and at some distance from it. The shilling remained unpaid up to the day of his death, and it was found that in his last will he had directed that his corpse should be carried by that lane to the place of interment.4
His will was dated 24 January 1754 and proved on 11 February 1766. Among other legacies, he left his wife Abigail "the life use and improvement of my negro man Caesar, my mulatto boy John, and negro woman Bett." Caesar had been purchased at about age 12, on 26 April 1721 in Barnstable, Massachusetts. He was admitted to the church in Wilton, Fairfield Co., Connecticut, in 1741, the only Black member, and accumulated enough personal property during his life to make a will in 1773.2,5
Moses and Abigail had the following known children: Abigail, Hannah, Abijah (b 19 November 1721, d June 1797), Martha (b abt 1723, d 4 August 1814), Moses (d 18 January 1789), Sarah (b abt 1726, d 1796), Phebe (b abt 1731, d 1817), Dinah.6,3
